Dual Luciferase Assay - (Apr/05/2013 )

Hello, I was wondering, when performing a dual luciferase miRNA assay, why do you only clone in the 3'UTR of the gene of interest and not the entire gene? Thank you!

I guess you are referring to miRNA target assay, right? Because miRNA targets (mostly) 3-UTR to inhibit protein translation or cause mRNA decay, a miRNA target is inserted into the 3UTR of the luciferase gene and the luciferase in the vector is used as readout of miRNA targeting effects.
