the storage time for primers - (Mar/26/2013 )

How long time can primer be stored in the freezer at - 20 C? I got my primers last July and worried about that if I can still use that to get the same sensitive result.

Without malfunction of the freezer or many freeze-thaw cycles, primers should be working even after many years. I use primers that I ordered 7 years ago without problems (actually diluted aliquots that I prepared at that time and froze too). If there's a change in sensitivity I don't know.

If they are in TE, they last essentially indefinitely, even at room temperature.

thanks. I will try one batch of my experiment tomorrow. I diluted primer in water.

I also store mine in water and it should be fine after this time. Can't you repeat a sample you used back then now so you can be absolutely sure about the sensitivity ?

I was using stock batches of primers many years old.
But also, one time the stock diluted only in water, kept in -20 all the time, thawed only few times.. got completely degraded.
I use only 10mM Tris pH8 to dilute primers since.

i wanna sequence my PCR product. Company person said that i need special primer for that. But i think my primer should be working for the sequencing.

Kindly help me with this.

Do i really need sequencing primer for that.

We reconstitute primers in TE buffer to 100 uM concentration for long term storage, and prepare 10 uM working solution using water.
