Cytotoxicity & LD 50 - (Feb/06/2013 )

Hi folks. I am doing a cytotoxicity assay on human lymphocytes. I am using two different test substances (of my choosing) and I am going to run my cells in a 96 well plate through a plate reader. This program will give me absorbacne values that i am supposed to put into excel and generate a graph. How do I get a lethal dose from there? Any help would be appreciated. Also, am I plotting absorbacne with time or some other parameter/variable?

OK - if you think about this a little bit, you will see the answer is pretty obvious.

To determine a lethal dose, what is it that has to change over your assay plate?

Yup, so it is the dose of drug that you have to change.

So my class doesn't have a multi well pipettes. Is there any advice/techniques to plate without causing contamination/pipettes errors?

Change tips between each well and look to check that you are sucking up (and dispensing) appropriate amounts of the things you are adding. You can also try reverse pipetting.
