How can i best delete a gene region of my host cell.? And how long time will it take?

You can use a recombination event using a plasmid which contains sequence found on either side of the desired KO region. Usually these sequences will also flank a reporter gene of some sort to help with identification of cells which have recombined.

Clonal selection of cells will take months! Make sure you are very good at your cell culture before you start so that you know what you are looking for, and so that you can check on the health of the cells, and be able to determine if the cells are behaving the same as the parent line (important if you need controls!).
