Brief vortexing for reconstitution of recombinant proteins? - (Sep/27/2012 )

Hello,

I have a very simple question.

Generally, vortexing is not recommended while reconsituting lyophilized recombinant proteins.

Can a brief vortexing (e.g. 10 sec. in 1xPBS supplemented with 0.1% BSA) result in a significant loss of the proteins' bioactivity? Did anyone experience problems because of vortexing?

Gunes.

vortexing can cause denaturation of the protein due to physical stress (and also, possibly, oxidation).

it can be performed if you can cause a swirling effect rather than bumping.
