can I dilute the drug with DMEM - (Aug/21/2012 )

hi there,
wishes, I am using a drug that should be dissolve in DMSO. I am working for to detect the cytotoxicity of this drug on mammalian cells by MTT assay. I have reffered from the research articles and researchers, that for doing this experiment DMSO concentration should be same or below of 0.1 %. So, may i dilute the drug by using DMEM from stock solution which is made up of DMSO.

yes you can; take care to add the same amount of DMSO in controls as well i.e. take pure DMSO and disolve it in the same amount of DMEM and then add on the control wells the same amount as you add for the drug. This would be the correct control to see only the effect of the drug not of the DMSO.

thank you very much for your kind reply and I will maintain the control DMSO as you suggested. but my senior suggested that if I dissolved the drug from DMSO stock in DMEM there is a chance of recrystallization may occur. so my question is if the chance of recrystallization occur when i dissolved in DMEM?

I once observed kind of a precipitate when I dissolved the DMSO drug to media but then I vortexed and it and it redissolved again. Anyhow, usually you are using far more media than DMSO so the crystals can be easily dissolved.
