How to release fixed nuclei from slide - (Aug/02/2012 )

Hi all,

Anyone have an idea how to fix nuclei to a glass slide, cover slip, or really anything else that would allow treatments (PNA hybridization, stringency washes) and then be able to release whole nuclei?

A standard fix by smear and drying should work. You could possuilby use something like gelatin or collagen to fix them on a slide. I suggest you ask a histologist or try for protocols.

Thanks for the reply bob. I got the things stuck with drying and I hit em with some cold methanol, but getting them back off without tearing them up is the issue. I'll surf histo-world and see what I net.
