can I use microplate reader for INF-gamma ELISPOT assay? - (Jul/12/2012 )

Hi, I pan to do ELISPT assay to detect the level of IFN-gamma released by PBMC under stimulation of certain pathogens.

Now I have difficuly in finding an ELISPOT machine.

I wonder if it can be replaced by other 96-well plate reader? We have a reader in lab that suitable for fluorescence and other non fluorescence light, and the temperature can be settled.

I don't quite undersdand the difference ot ELISPOT machines compared to other microplate readers.

Would you please give me some suggestions? Thanks a lot!

It sounds like you can, provided you have the appropriate filter(s) for your detection.

Besides, it supposed to be read mannually under microscope. I wonder if it needs special type of microscope, or it doesn't matter?
