Left my media at room temperature for 48 hours; okay to use? - (Jun/29/2012 )

Hi all,
I was in a hurry the other day and left my media out at room temperature for two days. It contains FBS, RPMI, L-glutamine, HEPES, and sodium-pyruvate. Should I make new media, or is it still okay to use?

It really depends on what you're using your media for. If it's for important experiment, you don't want this small 'change' to affect your whole/final outcome (i.e. what you're going to use your cells for at the end).

48h technically should be fine at RT, but depends on what your RT. 48h might be pushing the limit as 24h might be just fine.

End of the day, if you want to be safe, get a new bottle and remember to keep it in the fridge next time

General rule - "If in doubt, throw it out". Medium contains a range of components that degrade quite quickly at room temperature which can be critical for cell growth. You should throw it out and get a fresh bottle.

I guess you can still use them, if your room-temperature is around 18-20"C, as in an air-conditioned lab...

of course, do bear in mind that some components may be slightly denatured. so i guess maybe you can increase or even double up the volume for culturing your cells... eg. instead of 3-5ml for a T25 flask, put maybe 6-10ml...
