0.2M phosphate buffer for IHC - (May/18/2012 )

We have been using 0.2M phosphate buffer for IHC (for washing and antibody incubation), but I have seen most protocols use 0.1M phosphate buffer or PBS for IHC.

Could this formulation compromise results? We do get some staining, but not much.

Potentially yes, increased salts makes the buffer more stringent, so you are less likely to get binding.
