Using serum as the sample in Western blot - (May/18/2012 )

Has anyone ever successfully used mouse serum as the sample in a Western blot? If so, what (if anything) did you use to dilute it?

I have run westerns with human serum before, my samples were diluted in 10mM PBS because I performed albumin depletion beforehand. I would think you could use ddH2O, loading buffer or even your SDS-PAGE running buffer (TGS for example), its going to be in that anyway when you run the gel.

Can anyone tell me which protein should choose for normalization. I am doing western blot of depleted human serum samples but the problem is reference protein. how i will quantify change in expression level of protein of interest?

Hi.. you can dilute it with water, PBS.
