ATPase enzyme assay !!!!! - (May/18/2012 )

Hi

I am planning to do ATPase enzyme assay for a protein, I am using only one concentration 1mM and I want to test the activity versus time, can I interpret my results in percentage from OD instead of using the conventional enzyme equations? also can I use Michaelis–Menten and how can I use it practically.

depends on what you want to show and whether you keep everything equal (same volumes, same amount of enzyme).

it also depends on the linearity of your detection method if you just want to compare ods.

I just want to show that there is activity in X enzym during several points of time. let us say that I need to test ATpase activity of 1mM enzyme every 0,10,20,....60 minutes. in this situation how can I interpret the result by percentage or special equation?

you can interpret by percentage if everything else is equal and the detection method is linear for the range of your assays.
