Plasmid with two similar bacterial ORIs - Can it be transformed into bacteria?? - (May/01/2012 )

Dear all,

I have an urgent question please:

If I clone a plasmid that will carry two identical bacterial origin of replication (ColE1).
Will it trigger incompatibility? Or in other words, would it be able to propagate in standard competent bacterial cells (i.e. DH5a)?

If not, I would need please a solution of how to propagate in bacteria a plasmid with two bacterial ORIs.

ColEI plasmid origins do not work when there is more than one on a single plasmid. A plasmid containing two distinct plasmid origins can, typically, propagate, although I'm sure there are cases that will fail.
