What's the difference between megnatic beads and agarose beads to IP His-tag - (Mar/28/2012 )

I 'm planning to pull-down the interacting proteins of my 6XHis-tagged protein with anti-His beads. But I can find a few agarose beads for His antibodies. What's the difference between magnatic and agarose beads?

The magnetic beads use a magnet to pull them out of suspension, which is gentler than centrifugation, they can also be centrifuged though. The agarose (sepharose) beads can only be pulled down by centrifugation.

If I'm about to do IP to look for interacting proteins. Is magnetic bead a good choice? I see very few agarose beads for His-tag but a lot magnetic and nickel NTA something which I never worked with before.

It will work fine. NiNTA is usually used for column based preparations.
