plasmid extraction - (Feb/21/2012 )

all pictures are the same, which one look better? and does the result ok ?

It could be your plasmid. We can not tell you definitively if it is your plasmid. We know nothing about the plasmid. What I can tell you is there can be 3 bands from a plasmid prep (nicked, linear, supercoiled). If those bands are in the vicinity of the correct size, that should be your plasmid. The ladder on the right is very faint, and the ladder to the left is not necessary.

thank you.. but how about the gel? i need also your comment about the gel.. is there the band? can you explain to me about the gel..what else i can modify ?

I think the gels are probably OK. It could be the photography that makes them tough to interpret. there are bands definately there. If they are the correct bands, well that is for you to determine.

half of the pictures have a strange background pattern. i assume they show up when increasing exposure to better see the faint bands. you should find a way to eliminate the background and reshoot the gel with the better of the long exposures.
