How to..? Fungistatic & fungicidal evaluation. - (Feb/14/2012 )

How to evaluate fungicidal and fungistatic effects of a plant extract? Im using the pourplate method with SDA. Organism is Microsporum canis and Candida albicans. 10 ml Leafextract will be added with 10ml SDA in plate and when it solidifies, fungal incoulum will be stabbed at the center. Help!

What is the plant and anticipated application - or is this just observational? Not sure diluted (50%) SDA will solidify so well and the assay at best will assess fungistatic activity, though I suppose you could cut the inoculated agar section and culture by enrichment.

this sounds suspiciously like coursework to me....try to set up a detailed protocol of your experiment (step by step) and try to answer for yourselfy why and what you are doing at every point of the experiment. Ask yourself what the results obtained at any point of your experiment will tell you about the antagonistic effects of your plant extract on your test fungi, and keep in mind that more than one step might be neccessary to get good and watertight results. Do not forget about control reactions - which you will need to compare your results to. There might be a need for more than one control as far as I can tell from the sparse information given which is not enough to give you any more specific advice for your exp.
