Possible non-specific staining of ribo-probe in Fluorescence in situ hybridisati - (Jan/01/2012 )

I am a little stuck on whether my ribo-probe has worked, I have used a dig-labelled probe with TSA amplification and previously it worked. However this time I used a probe to label PRDM-16 mRNA and got what seemed to be nuclear staining (as it co-stains with DAPI in brown fat). I know that as a protein it is expressed in the nucleus, but surely as mRNA it should be transported to the cytoplasm.... and so stain there? I haven't made a sense control probe to compare it to as I ran out of time... Any idea
