Potato tissue culture pH - (Dec/14/2011 )

Hello, one of my projects in grad school these days involves a lot of tissue culture of potatoes, and today I made up a batch of MS (plus vitamins) media with phytogel, but I was in a hurry, and I forgot to pH it to 5.6 like I usually do...

My question is: can I still use it? or is it going to have problems growing my little potato friends? It seems to have set just fine, but before I throw it out and make new, I thought I'd get a second opinion.

what is the unadjusted pH? if it's not near 5.6 then you must either adjust it or prepare it fresh.

I believe it is typically around 5.

you can take a chance but you would be better off adjusting it or preparing fresh.

I think I am going to do both. I'll put a few cuttings on the un-pH'd and make new as well. That way, I'll know the consequences if I ever make such a mistake again. Thanks for your replies.

no problem with that re adjust the pH and just add the compounds that is not autoclavable such as Antibiotics and some of the Vitamins,
but I am not sure that you could melt and re gel, phytogel.
one more problem is with pH meter! they usually work best at 20 degree centigrade!
