Broader lanes on SDS PAGE - (Oct/31/2011 )
Dear friendz,
i am getting broad lanes and diffused(not sharp) bands on SDS PAGE when running histone proteins. i run my sample at 100V for 2hrs on 12% gel with 10% APS. Plz tell where i am making a mistake

hey Binsan....a picture and more details about your PAGE/western protocol would help........is this a mini-gel bec running for 2 hours at 100v seem long....

this can be caused by buffer components (of the protein sample without page sample buffer).
in what buffer is your protein?
are there other proteins in your sample that behave normally?
