dear friends can anyone recommend me a basic protocol followed in western blotting and in plant dna isolation

For the Plant DNA extraction, there are many different protocols, depending on the type of plant material you are using. The woodiness of the plant and the source material (leaf vs stem, root) will make a difference, so I would recommend doing a more specific literature search with the exact plant and source material. That said, most plant DNA extraction protocols use a chemical called CTAB as well as PVP. I have posted a link to one such protocol:
