Quantifying DNA from Buccal Swabs - (Oct/13/2011 )

So I'm collecting DNA from Buccal Swabs but having some trouble quantifying it. Is there any DNA extraction kit that allows the use of Nanodrop???
Otherwise, Im trying to create a standard curve for fluorescence using Hoechst Dye. Right now I've got DNA concentrations between 0 and 2.6 micrograms/ml and 1 mg/ml Hoechst Dye in a 100microliter well with buffer.
352nm excitation, 461nm emission wavelengths...

But my values are all over the board and not linear at all.
Any idea where I could be going wrong?

I'd make sure my quantitation was working by testing serial dilutions of a known DNA concentration. Those concentrations sound very low, and are likely near the limit of detection, but a serial dilution test will tell you if that is true.
