ISH probe choose - (Sep/05/2011 )

Hello everyone!

I'm new with ISH and I have a doubt... I'm styding vascular tissue in plants and I would like to know how many different probes should we use... Is only one probe ok?? Our should I use more than one??

I have some protocols, but I can't get this information anywhere!

You should have a number of probes that are used routinely for your experiments, the typical ones are Specific probe, anti-sense probe (i.e. opposite sequence to the probe - identical to target sequence), scrambled probe (specific probe sequence scrambled or with same G/C content). These act as controls to ensure that your probe is binding to what you say it is.
