Putting the cells on ice is the best way to (mostly) stop cellular functions. You can rinse in cold PBS and use cold lysis buffer to make sure that there is the lowest possible amount of activity.

Whether you go for a breadth of compounds or focus on a smaller number is a discussion you should have with your supervisor/boss. It is definitely a good idea to titrate the effects out and use amounts that you know work for your system. Just don't make the mistake of assuming that something that works in one cell line will also work in another, even though that may well be the case.
