how to determine a plant promoter activity in bacteria - (Aug/20/2011 )

Yes, i agree with you.The thing , want to do is: I have a plant beta glucosidase gene promoter sequence. I want to test if it gives promoter activity. But i can't test it with transgenic plants in my lab.So, I want to use yeast expression system if possible. Despite all my search, i could't find a kit proper for this. Isn't there any study before like mine? I couldn't find it either:(

Hmm first maybe you should find the system for yeast and do transfection once you got one...but based on what i know, normally they already contained promoter in the plasmid...otherwise you need to get the backbone that suit for yeast transformation and construct all by yourself...it will takes time, try to search transformation in yeast let see what you will get...I guess transformation in yeast is not rare, so you will have lot of method just in different strategy..Do you really need to use that promoter? i mean if you cant try it directly on plant it's hard to know the effectiveness of the pomoter right? maybe if you can do transgenic plant, agrobacterium can help you :) but for me yeast can work too...as long as you can get suitable system for (plasmid) once you insert Promoter+Gene then just use electroporation ...:) hope it works..
