Does tritonX100 disturbs sds page? - (Jul/27/2011 )

Hi everyone!

does anyone of you know, whether triton x100 influences the running behavior of sds samples? and if yes, how can i get rid of it?

yup, triton can compete with or displace the SDS in the sample buffer so it may interfere with the mobility of your protein or cause distorted lanes etc....but I guess it depends on how much you use for lysing or extraction. We use it at a final concentration of 0.1% (0.2% + 2x Laemli buffer) and we don't have any problems. I suppose if you use 10x more concentrated, you may have issues... then you can try some highway robbery clean up kits or resins (Pierce etc) which will bind the detergent and you elute the proteins or vice versa, they bind the proteins and you can wash off the detergent. And Sigma has beads I think though I'm not sure. I think dialysis will not work for triton but you have to check this out...
