his tag purification - Nickel vs other resins (Jul/19/2011 )

Hi,
I have been using Ni-NTA for a while now for my specific protein. But still either I don't get enough purity & sometimes yeild is very poor.
I want to hear other user experiences if someone has tried resins other than Ni-nta on the market. I came across His60 which is Nickel based resin & also another called Talon. Has anyone had any experience with these two?

The other one was Talon, seems it is cobalt based. So the question is does Cobalt really gives higher purity than Nickel ?

Hola, I have used both in batch and Nickel in colum , moreover any of suppliers have different capacity charged resins. For me is quasi impossible have your protein pure after one sole purification step and to have more pure protein you have to choice enriched fractions and perform a ion exchanger and/or gel filtration. different thing is about low yield, does it mean that you have protein in the flowthrought? this could be due to different causes and you have optimize your process. Wait for more opinions and buena suerte
