IC50 calculation software (need help) - (Jun/01/2011 )
hi all, can some one suggest a software for IC50 calculations (free oder paid) and please share some experience of the same. Will be a great help for me
try GraphPad prims
it is easy to use
choose the xy table+ graph
them use your x valeus log (10)
analise with the non linear fit curve log (agonist)vs response
hope that helps
also see the topic http://www.protocol-online.org/forums/topic/8569-how-to-calc-ic50/ they have good sugestions
u can also try
MasterPlex ReaderFit 2010
i never used that program but it seems easy to use
you can see the video
If you have someone to show you how to do non-linear regression you can set up the calculations on MSexcel (or any other spreadsheet). Not as refined as graphpad but the results are the same

You should never use Excel for IC50 estimation. Your viability data is not linear or fits simple log transformation. For accurate calculation of IC50, a nonlinear curve fitting program that uses the Four Parameter Model is needed.
A paid program such as GraphPad Prims is great, but there is a free online program BioDataFit, which produces the same
To learn how to prepare the raw data for inputting into an IC50 program, read here. To learn what to do if your experimental data does not fit the Four Parameter Model, which can be very
Good luck and hope you all will smoothly get your cytotoxicity experiments done and get publication quality IC50s from these tips.

This should read “should never use Excel for IC50 estimation if you are unsure of what you are doing”. I haven’t come across anything in Graphpad that can’t be done in Excel or libreoffice. Graphpad is just a spreadsheet with less flexibility.
Haha... DRT may be right. Or it should read “should never use Excel for IC50 estimation if you can't write a macro that incorporate the four parameter model for nonlinear fitting of a sigmoid dose-response curve.”
DRT, we would be happy to try your Excel IC50 macro if you would post it up, and compare it to BioDataFit, in case BioDataFit goes offline.
