MFI and microscopic visualization - (May/31/2011 )

Does anyone know how MFI from flow correlates to the ability to visualize cells on fluorescence microscopy? That is, if I have a population of cells that express GFP, what minimum MFI would I need on flow to be able to see the cells under a fluorescent microscope? Thanks.

This is all relative to equipment settings, including power and gain settings on your FACS machine and lamp/laser power settings on your scopes. The way we correlate this is to either buy from BD/Invitrogen some standard beads with different, known amounts of fluorescence and take some scope images of them in dishes and run the same beads through the FACS machine. You can then take those FACS settings and run your cells to see if your population falls within the limit of detection based on their MFI relative to the beads and your scope image data.
