Stripping Buffers that won't kill my cells? - Recipe for 0.1 M Tris-Acetate 2.0 M NaCl pH 7.7 (May/06/2011 )
Does anyone have the recipe for 0.1 M Tris-Acetate 2.0 M NaCl pH 7.7 (Pierce Biotechnology, Inc.)? I need this stripping buffer to take the antibodies off of my target cells without killing them.
Does anybody know of any other stripping buffers that will take off the Ab without killing cells?
Thank you so much in advance!
Note: I don't actually know this buffer, but the standard method is...
Calculate and weigh out the amounts of tris, and NaCl for your required volume - for 1 litre, tris will be 12.14 g, NaCl will be 116.88 g (you shoudl check these for yourself though as I am doing this off the top of my head). pH to 7.7 with glacial acetic acid.
Also note that this buffer will be strongly hypotonic so it will kill your cells by dehydration if left on for too long.

(here comes the drive-by nitpicker) that's hypertonic
Whoops, quite right...
