Sybr Green Real Time PCR - Melt curve - Any problem on the melt curve? (Apr/08/2011 )

Dear all,

Here is attached the melt curves of my SYBR Green Real Time PCR. I have run all the PCR products on agarose gel. All got single band. However, I'm not sure if these results are valid since some melt curves shown one major peak and also one or two very small peak(s). Any comment?

Yes, your result are valid, you have almost perfect melting curves. This is how melting curve should look like.

I agree with vladooo, very nice melt curves. In addition to melt curves though, you need amplification plots. As long as your amplification look like nice sigmoidal curves, you can assume your assay are working the way they should.
