Sodium Butyrate Stock Preparation - pH; histone preparation; acetylated histone (Apr/05/2011 )

I got sodium butyrate crystal for acetylated histone preparation. The protocol I got requires the sodium butyrate stock (500mM) with pH 8. And the protocols I found from online is to use NaOH to adjust pH value, which doesn't make sense because the pH value of sodium butyrate solution is higher than 10 already. My understanding is to use NaOH adjust butyrate acid. Since we don't have butyrate acid in lab and I already got sodium butyrate, can I use HCl or other acids to adjust the pH value? Or I just don't need to bother the pH? Thanks a lot!

what other components are in the buffer? that can help you determine what counter ion (acid) will be safe for reducing the pH.

Thank you very much for replying. I added sodium butyrate to TEB buffer (Triton X-100 0.5%; PMSF 2 mM; NaN3 0.02%; NaBu 5mM; made with PBS) for acetylated histone purification. Since NaBu is 1:100 diluted in the TEB buffer, I just added it directly to TEB without adjust the pH value of stock. It turned out that there almost no effect to the pH value of TEB buffer. So I don't bother to adjust the pH value of NaBu stock now. Thank you again!
