Can I use delta delta Ct? - (Apr/05/2011 )

Hi,

I should begin to deal with methylation qPCR. I want to quantificate methylation status of P16. I would like to know if delta delta Ct method is appropriate for such analysis. In papers that I am reading, authors mention standard curve analysis ( relative or absolute), but not delta delta Ct.

hi, I cant't help you with that, I had readed in the most papers they use a standard curve, do you know how to calculate the ratio between the methylation fo your target gen and beta actine gen?, they use the log of the quantity?, they use the quantity and then they apply the log?, how can I analyse that?, I don't speak english, so sorry for my grammar or spelling errors.
