Plaque reduction neutralization test (PRNT) - (Mar/24/2011 )

Hi Everybody
I'm looking for help, I want to do PRNT for detect past infection with Dengue viruses, I have stock titer of D2 5 x 10⁶ PFU/ml.
I want do serial dilution from stock to get 800 PFU/ml. Please how i can prepare this titer from my stock?.

you could for example start by adding 1 ml of your stock solution with 4ml of whatever you use to dilute so you go from 5 x 10^6 to 1 x 10^6.

And then you can start doing 1/10, 1/100 or 1/1000 dilutions till you reach 1000 PFU/ml and in your last step then you can make a 800PFU/ml stock solution.

another example is:


take 1 ml of your stock (5 x 10^6PFU/ml or 50 x 10^5 PFU/m) and add 5,25 ml ===> you will then have 50 x 10^5 PFU/ 6,25 ML or 8 x 10^5 PFU/Ml .. and from there you can dilute in steps of 1/10 or 1/100 or 1/1000 till you reach 800PFU/ml ...
