3 distinct bands of gDNA on gel - (Mar/23/2011 )

Hi,

I was wondering if anyone could help me with my problem of getting three distinct bands of the gDNA when run on gel. I extracted the DNA from gram negative bacteria and got three distinct bands of 5 kb, 6 kb and 12 kb, when run on the gel. The gDNA which is around 21 kb is not visible. I am attaching the image of the gel.Attached Image

Likely your organism has multiple plasmids, but possibly fewer with alternative supercoiling. You can gel isolate the bands and try restriction mapping them, or try transforming them into a strain missing the plasmids.
