effect on tris on yeast growth - (Mar/15/2011 )

hi
I want to grow yeast with a salt at neutral (and constant) pH and hence i used tris-cl buffer for the same. But now i have come to know that tris deactivates some enzymes....so i just wanted to know is it beneficial to continue the same experiment ??? or does tris have any other effects on yeast growth ??? Or is there any other buffer (pH 7 ) which have no side effects on yeast growth ???

Hola, it seems that you want grow yeast in a flask,because in fermenters there is a pH control, so if you use any sugar the liberated CO2 will acidifie the medium, if no sugars deamination of aminoacids will increase pH.So, itīs more adecuated a phosphate buffer pH7 and the molarity will varies in function of the nutrients concentration, for instance for 1g/l of sugar I will use 50mM increasing untill a maximun of 0.2M for bigger amounts, but I donīt know how calculate it "a priori" so you will have to do a try.
