Primary AB over night at room temp? Screwed? - (Feb/17/2011 )

tonight i was staining slides of pfa fixed tissue, i put on primary antibody for PECAM in PBS with 10% donkey serum and 2% Tween 20. I left in a hurry and the slides are at room temp over night with primary... instead of in the cold room at 4 degrees... will this be a major problem? am I screwed?

The slides are sealed in a humidity chamber if that helps...

as long as they didn't evaporate to dryness they should be fine. antibodies normally survive and operate at body temperatures.

although, you may have to repeat the conditions if the results come out well.

My experience - some of the Abs I use are not effective during overnight incubation at fridge, but give very good reaction at room temperature. I have repeated this several times and got the same result. If the slides have not dried I don`t see problem.
