Optimization for cell viability assay (MTT assay) - (Feb/01/2011 )

Hi. Im doing a study on effects of certain extracts on cells.

Im using mcf-7 and chang liver cells. im in the optimization stage now. (Im using the MTT assay to determine cell viability)

i did a trpicate for each concentration of cells. but the three reading are not so consistent , for instance one give 1.0 , while the other two reading gives 0.8 .

moreover , a higher reading is observered for 5000 cells compared to 10,000 cells.

im carcking my head over this !

Any suggestion ???

i MA HAVING TROUBLE SATINING THE pbmc FOR FACS. CAN ANY ONE PLEASE TELL ME HOW TO IMPROVE THE FACS STAINING AS ALL MY CELLS DONOT TAKE UP THE CONJUGATE AND I END UP ONLY A SMALL SHIFT OF THE OEAK
