Calculating - RNA extraction (Dec/14/2010 )

Good Day,

In Bugos et al. (1995)article, he extracted RNA as follow:
Tissue was ground to a fine powder in liquid nitrogen and added 1:2 (w/v) to 25:24:1
phenol:chloroform:isoamyl alchohol and vortexed.


My question is:
If i started with 0.1 g from plant tissue (leaf) how much i should add from phenol:chloroform:isoamyl alchohol?

0.2L or 0.2ml or what from P:C:I?

if 200ml, it's consider too high. isn't it?

He used 1:2 weight per volume - as you have 0.1 g use 0.2 ml.

0.2ml not 0.2L?
but in calculating normally we used gram with litter, so when I used 0.1g then I should use 0.2L (200ml)
is it true?, or up to us, we can chose any unite as we want?

Please can you illustrate a littel bit more?

I think you might be confusing using molecular weight in calculations (i.e. g/L) with ratios.

In ratios weight and volume need to be equivalant measurements:
kg=L
g=mL
mg=uL
