promoter isolation in Aegilops - (Nov/12/2010 )

I want to isolate a gene promoter in Aegilops (the wheat ancestor)that I only have a partial cds (156 nucleotides)of it. As Aegilops is not sequenced completely yet,I need a suggestion for isolating the promoter of the gene of interest.
I should mention that I have the sequence of my gene and its promoter in rice(oryza sativa),sorghum and corn(zea maize).

How far away is the promoter? You can get a few thousand surrounding bases pretty easily with inverse PCR, but it will be difficult to get, say, 50 Kb. Another approach would be a cosmid library screen using the sequence you have as a probe. Someone (or you) may already have such a library.
