Pediocin resistance protocol - (Nov/08/2010 )

Afternoon all,

I was wondering if anybody could help me. I am looking for a standardised protocol to assess the development of pediocin resistance in certain bacteria. The papers I have read have all favoured cell counts on specific concentrations of pediocin on agar plates but they all seem a bit wishy washy and dont fully elaborate on the protocol (that may just be me being over cynical!!)

Feel free to insert *antibiotics* where I have pediocin cos I guess its essentially the same idea. Any help or ideas would be greatly appreciated.

Hola, I will seed a sterile 96 well flat botton plate with 100-200 ul of a fresh inoculated culture and in each column pur the dilutions of pediocin and incubate at the selected temperature., following the OD changes in a plate reader at differen times. In this way I could know wich is the adecuate concentration to develope the resistence. Do you think it could be a fiable method?. Good luck
