nuclear and cytoplasmic RNA - (Nov/05/2010 )

Hi everybody,

I am suspecting that i have immatur RNA transcripts (PremRNA) in some cells line's cytoplasm.
I thought about separating the nucleus from the cytoplasm and extract RNA from each. Then, i want to sythesize cDNA and check if i have premRNA expression in the cytoplasm and compare it with the expression of premRNA in the nucleus.

Any body knows any way to separate the cytoplasm from the nucleus and extract RNA from each separately?

You can use Qiagen RNeasy kits with some modifications.
