How to homogenise tissue in buffer? - (Sep/29/2010 )

I grind 30mg mosquitoes (~15-20 individuals) in a 1.5ml tube with a micropestle after adding in Liquid nitrogen and continue to grind after adding buffer. Does this considered homogenising? Or i'll had to use syringe?

If i am to use a syringe, how do i load the solution into it? By taking it through the tip? Loading with a pipette? How does using syringe differ from using a pipette to mix the solution?

There is some sort of automater tissue grinder/homogeniser (i am not sure whether it is a rotor stator) in my lab. If i use that to grind my 1st sample, how should i wash it for my subsequent sample? Can i use ethanol?

grinding in a mortar and pestle (1.5 ml tube and micropestle) is considered homogenization (for the purpose of extraction).
