miRNAs and antibiotics - (Sep/01/2010 )

Hi,

I have transfected miRNAs into N2A cells. The goal is to look at how the expression of certain proteins are affected by these miRNAs. However, normally, I do these experiments without any antibiotics in the medium. But as there are a lot of people working around me, who have contaminations, I would like to add PEN/STREP to my medium. Now here comes the question: I was wondering if addition of antibiotics would affect the result (compared to experiments without antibiotics).

Concerning your results you should be safe: using antibiotics in both transfected and mock cells you'd have the same "miRNA background". considering your question: yes, it could certainly be that antibiotic treatment could affect miRNA expression (like any stimulus, e.g. transfection). maybe you'll tell us for sure!!! :P
