PCR with one primer - (Aug/31/2010 )
I came across this paper published in 1992, where the author has used only the forward primer for his PCR. He has mentioned a range for PCR product with each of the primers used.
My question is, if I have to repeat this experiment, how can I exercise control over the size of PCR product and make it consistent? or is it that the length of the product will be dependent on extension times alone???
as a rule of thumb 1 min elongation at 72°C is needed for a product of 1000 bp length, but not sure if this is of any help for your experiment.
