Retrovirus production - How to produce retrovirus with Transient transfection? (Aug/27/2010 )

Hi there!

I was producing lentivirus using the pLKO system (pLKO, pspax2, PMD2G).
Now, I want to know how to produce retrovirus without a packaging cell line. Which plasmids should I use to get the retrovirus? I have some construsts in pBABE, and I guess I can use the PMD2G (VSV-G), but what other plasmid do I need?(because the pspax2 is for lentivirus)

However, I will recommend using packaging cell line for retroviral production.

Thanks a lot. I actually had platinum E and phoenix ampho, but they were not properly frozen, so now I have to wait for the new cells to arrive, and that is why I was checking whether I could produce viruses with transient transfection in the meantime. But it looks like I dont have the plasmids anyway, so I will wait for the cells.
