Long term storage of bacteria in fetal calf serum -70 or -80 - Temperature issues (Aug/24/2010 )
Hi
May I ask which temperature is most optimal for keep bacteria in fetal calf serum. -70 or -80? I did observe the usual temperature for storage of bacteria is -70 degree celcius... But due to the original factory setting in my freezer I have been using -80 for a long time. I am paranoid whether this is acceptable or whether this will have any effects on my isolates??

-80 is a temperature that is often used. Especially in stocking cultures in liquid nitrogen.

That would not be different between -70 or -80. I think glycerol is needed in the preservation of bacteria.

I store my bacteria at -80 in glycerol - 500 ul 65% glycerol + 500 ul bacterial culture.
