Deactivation of RBCs from Whole Blood? - (Aug/12/2010 )

I was wondering if there was any way to deactivate RBC's from whole blood and not effect any other cell types significantly. I would try lyse buffer with Ammonium Chloride but I was thinking more of blocking RBC receptors or something similar. If I had to lyse I would but again, I dont want to damage anything else significantly other than that RBCs. Thanks!

Hello :) There are commercially available RBC lysis solutions - I make a home made version, but I have the recipe at work. I use it to get rid of RBC in whole blood and BM.
If no one else posts a recipe tonight, I'll post it tomorrow :D

here you go: my RBC lysis solution
0.2M NH4Cl
0.01M NaHCO3
0.13mM EDTA

I would usually resuspend the cells in the RBC lysis solution, leave it for a few minutes on ice. Then, either spin down the cells and resuspend in fresh PBS etc. Or add some PBS before spinning down.
