Chromosome Specific Enzymes - Mammalian enzymes that cut only one chromosome in genome (Aug/12/2010 )

Hello,

I have an assay where I digest all chromosomes with two generic primers and then place the digested samples on a Southern blot for visualization to look at size differences. However, I am unable to identify which chromosomes have a particular size. Are there any enzymes that are specific for individual chromosomes, such that they cut only one chromosome and not any other? (i.e. - a single cutter)

Southern blots do not give chromosomal localisation, you need to do FISH (fluorescent in situ hybridisation) or a similar technique. The probes you use for the Southern should work for this.
