Hi there,
I am hoping that someone can help to check that my dilution strategy is ok. I am starting in vitro experiments on a plant based protein soon and still am confused about the dilution strategy. I have been taking 1mg of this compound and diluting it into 1ml of my culture medium buffer. This would give me a 1mg/ml stock solution.
I was than going to add into well plates, buffer medium and different amounts of my stock solution to give me varying concentrations. For example I am hoping to have 50uL, 100uL, 250uL, 500uL and 1000uL exposure concentrations. To do this I would take ie: for the 1000uL concentration, 10uL of my stock solution and add this to 90uL of buffer medium- is that right? And for the 500uL concentration I would take 5uL of stock solution and add it to 95uL of buffer medium.
If any one can help me with this it would be very much appreciated!

1mg/ml = 1000µg/ml = 1000µg/1000µl or 1 µg/µl
if you take 10µl of this stock solution and add this to 90µl buffer, then how do you hope to have 1000µl in that medium?
I do not understand this...
And to be honest, I do not understand why you work with µl ?
Normally when you make such a dilution or experiment the aim is to have (for example) 10µg of substance X per plate, 1µg per plate, ....

Yeap. You are listing volumes there and not concentrations, so it's kind of hard to know what you are asking.
