Immunology - (Jul/14/2010 )

Dear all,

I am trying to express a protein in Dendritic cells and see the immune responses. I am failing to see the immune response. I would like to determine whether the protein is getting expressed or not. I could detect the mRNA by RT-Q-PCR. Now, I would like to know if the protein is being processed and being presented. Is there a way to show this? Can anyone please help?

Thanking you in advance.

If you have or can get antibodies against your protein of interest, you can use flow cytometry, immunocytochemistry, immunofluorescence, Westernblotting or even ELISA.

If you don't and can't have antibodies but your protein is characterized, you can try proteomic approach - eg. LC-MS/MS or 2D PAGE followed by MS.
